These naturally complex social behaviors are composed of innate and learned components that ultimately influence animal reproductive fitness. As aninnate behavior, the neural basis underlying aggression is genetically hardwired. However, cognitive abilities, external sensory information, past experiences and the internal state of the animals also strongly influence the display of aggressive behavioral patterns, demonstrating the adaptive and malleable features of this complex behavior(1, 2). The ecological and ethological significance of aggressive behavior (species-typical aggression), however, has to be distinguished from the maladaptive one (hyper-aggression). Pathological forms of aggression have long been investigated in the context of psychology influences. Decades ofstudies have demonstrated that both environmental and genetic factors influence aggression, however, the roots driving hyper-aggressive behavior remains poorly understood.In this study, we will use Drosophila melanogasteras a model organism to investigate the impact of hyperaggressivity on other social behaviors. Like other animals, in competition for food, mates and territory, both Drosophila males and females exhibit aggressive behaviors in same sexpairings(3).However, only male flies establish dominance relationships between competitors, while female fights end up with flies sharing resources(4). Selection of wild type males for their fighting abilities for generations yielded to hyper-aggressive flies, called “bullies”, with specific phenotypes: i) they employ extreme fighting behavioral patterns, ii) they do not establish dominance relationship because of retaliation, iii) only males are hyper-aggressive, and iv) they have lower reproduction success rate(5). To date, some candidate genes have been associated with hyper-aggressive behavior(6), however, we know very little about the link between hyperaggressivity and fitness.Pioneering studies showed that dominant males had a reproductive advantage, however, other studies that selected animals for their hyper-aggressive behavioral trait have shownthat these males were less likely to reproduce when competing with some"control"animals(5). Our preliminary results confirmed these previous observations. Thus, the objectivesof this internship is to better 2understand the link between hyper-aggressive behavior and reproductive success. To tackle this question, we willperformbehavioral tests (aggression and reproduction assays) using hyper-aggressive and control animals in order to confirm the previous observations. Then,we will manipulate a) the expression of specific genes (known to be differentially expressed between hyper-aggressive and normal animals(6)) and b) the synaptic transmission of neuronswithin particular brain areaswith thermosensitive genetic tools during Drosophila aggression and courtship assays and measure the behavioral consequencesin various experimental conditions. The aim is to identifythe brain structures and molecular mechanisms involved in the regulation of reproductive behavior. The fundamental aim is to understand the relationship between hyperaggressivity and the innate reproductive behavior in order to identify the regulatory mechanisms.During this internship, the studentwill learn to 1) work with Drosophila as a modelanimal, 2) performbehavioral tests (male-male aggression and courtship assays withmales and females) under different environmental conditions, 3) observe and quantify the behavioral responses (automatic analysis via MATLAB and manual analysis for specific behaviors) and 4) interpret the results obtained and draw conclusions.References1.Y. Exploration de la capacité de discrimination de nombres par les drosophiles

Projet: Savoir évaluer des quantités est une capacité utile dans de nombreuses situations, pour choisir la meilleure source de nourriture ou évaluer le rapport de force dans un contexte de compétition par exemple (Nieder, 2020). Cette capacité semble d’ailleurs très largement répandue au sein des Vertébrés, des poissons aux primates (Agrillo & Beran, 2013).

Au sein des Invertébrés, des études pionnières chez les abeilles ont démontré une capacité à dénombrer un nombre de repères visuels à franchir avant d’atteindre une source de nourriture ou à reconnaître un nombre donné d’objets dans une image (Chittka et al, 1995 ; Dacke et al, 2008 ; Gross et al, 2009). Dans ces deux cas, les abeilles peuvent reconnaître un même nombre indépendamment des propriétés des objets ou autres caractéristiques des images comme la densité ou la surface cumulée des objets. Cette capacité d’abstraction du nombre est en effet indispensable pour conclure sur l’existence d’un sens du nombre. Cette découverte inattendue chez un insecte interroge sur une potentielle universalité des mécanismes sous-jacents et sur les conditions évolutives permettant l’émergence d’un sens du nombre. Il s’avère donc nécessaire d’élargir nos connaissances sur la perception des nombres chez d’autres invertébrés pour répondre à ces questions fondamentales.

La drosophile est un animal modèle idéal pour l’étude des comportements pour plusieurs raisons : - l’existence de tests comportementaux robustes et faciles à mettre en place ; - un cerveau simplifié par rapport aux mammifères mais capable de capacités cognitives générant des comportements complexes ainsi que différentes formes de mémoires ; - un nombre très élevé d’outils génétiques permettant de manipuler l’expression de gènes et l’activité neuronale. Ainsi, révéler une capacité à appréhender des quantités chez cet insecte non social serait une véritable découverte et ouvrirait la voie à la compréhension des mécanismes neurobiologiques sous-jacents.

L’objectif de ce stage sera donc de tester la capacité des drosophiles à différencier des stimuli visuels sur la base du nombre d’objets présentés, dans un protocole d’apprentissage discriminatif à renforcement alimentaire. En cas de succès, nous chercherons à identifier les zones cérébrales impliquées dans la reconnaissance des nombres grâce à l’utilisation de transgènes thermosensibles permettant l’inhibition ou la suractivation neuronale uniquement durant les tests comportementaux.

Basic Function and Responsibility: 

The Postdoctoral Associate will lead research on the subject area outlined in the research statement submitted as part of their competitive application. Rose Postdoctoral Associates work under the supervision and mentorship of one or more senior Cornell scholars, while having substantial agency and opportunity to pursue their own research program. No formal teaching is expected, but teaching opportunities that benefit the career development of the Rose Fellow can often be arranged.

  • Experience with grant writing and budget development helpful.

Associate Professor Faculty Position

 The Department of Pathology at the Wake Forest School of Medicine (WFSM) invites applications for the position of Associate Professor (tenured or tenure eligible) in the Section on Comparative Medicine. The qualified candidate is expected to have a nationally-recognized, extramurally funded, research program. Experience and/or interest in nonhuman primate models is highly desired. This position comes with a competitive start-up package and excellent collaborative opportunities with both basic science and clinical faculty in the WFSM.

 

The successful candidate will become part of a multidisciplinary team of scientists in Comparative Medicine who conduct translational research using nonhuman primates, rodents and cell-based models. Comparative Medicine is home to 16 PhD, DVM, DVM/PhD, and MD faculty. Areas of research strength and growth in the Section include cardiovascular disease, metabolic disease/cell stress, behavioral neurobiology, radiation effects, cancer, and pediatric/developmental programming.  NIH-funded training grants within the Section support research training for postdoctoral fellows and veterinary student summer fellows. The Section is home to two NIH-funded national resource nonhuman primate (NHP) colonies (the Vervet Research Colony consisting of over 300 genotyped and pedigreed vervet monkeys, and the NIAID Radiation Survivor Cohort, consisting of over 150 rhesus monkeys previously exposed to radiation) with a total population of approximately 500 NHPs. Additional onsite resources available to faculty include imaging capabilities for large animals (CT/DEXA) with expansion underway to include PET/CT, MRI and a linear accelerator onsite (currently available at a separate campus), and a ~2600sqft ABSL-3 facility. These primate resources provide for abundant research opportunities and serve as magnets for collaborative work.

 

Institutional centers involved in collaborative research include the Clinical and Translational Sciences Institute (CTSI), the Cardiovascular Sciences Center, the Wake Forest Institute for Regenerative Medicine, the Wake Forest University Comprehensive Cancer Center, the Center for Precision Medicine, the Sticht Center for Healthy Aging and Alzheimer’s Prevention, and the Translational Imaging Program. A few of the many CTSI programs designed to support new research faculty include a Translational Research Academy, K Award Writer Series, Research Studios, and the opportunity to apply for a 2-year KL2 Career Development Award. The broader community consists of over 1000 faculty spanning over 40 academic departments and over 25 academic Centers or Institutes.

QUALIFICATIONS

The qualified candidate is expected to have a nationally-recognized research program supported by extramural funding and will have an outstanding record of research accomplishments and publications. Candidates should hold a terminal degree (DVM, PhD, DO, Pharm D or MD), and have a strong publication record. Postdoctoral research training, strong quantitative, oral and written communication skills, familiarity with animal and cellular imaging modalities, molecular and cellular pathology experience, and complex biomolecular and biostatistical analyses will be considered strengths.

 

RESPONSIBILITIES   

 

Successful candidates will be expected to maintain an extramurally funded research program. Support of institutional and departmental teaching and mentoring for graduate students and postdoctoral fellows is essential. Collaborative research with other faculty in the department and across Wake Forest Baptist Health and other universities in North Carolina is expected.
